Basic Calculations, Protein Function

Note:
This homework will not be collected. However, quizzes and exams will assume that you have completed and understand the homework assignment and could answer related questions.

Reading Assignment: Chapter 7 of Biochemistry.
Study Exercises: Chapter 7, p. 194, #1-5, 8-9
Note: Show all work and remember to incorporate your units in your calculations.
  1. For your Senior Honor's Thesis, you need to make up a 100 ml 40% stock solution of acrylamide:bis acrylamide [29:1]. Your advisor explains to you that the 40% solution contains a ratio of 29 grams of acrylamide to 1 gram of bis acrylamide. You show your advisor that the MW of acrylamide is 71.08 g/mole and bis acrylamide is 154.17 g/mole. She gives you a hint: A 1% solution is equal to 1 g/100 ml. Calculate how you would make this 40% acrylamide: bis acrylamide [29:1] solution.

  2. The protein that you are purifying for your Senior Honor's Thesis is about 50,000 kDa. Thus, you estimate that you need an 8% acrylamide gel to separate it from other proteins. Calculate how much of the 40% acrylamide solution that you made above in problem #1 would be needed to make 20 ml of an 8% acrylamide solution.

  3. Draw tryptophan, tyrosine, and phenylalanine. What are each of these amino acid's three letter code? What are each of these amino acid's one letter code?

  4. Draw the complete chemical structures of the peptide Pro-Arg-Lys and also the peptide Ile-Pro-Val.
What is the one letter codes for each of these peptides?

  5. Who won the 1999 Nobel Prize in Medicine? What discovery did this person or people make that it is worthy of a Nobel Prize? How is this discovery related to what we are currently studying in Biochemistry 3304?
